RESUMO

Hydrothermal oxidation was used to prepare a nickel-titanium alloy ( NiTi ) solid-phase microextraction fiber. The experimental results demonstrated that a nanoporous NiTi oxide composite coating was in situ grown on the surface of NiTi substrate by direct oxidation in aqueous H2 O2 solution at 80℃. The resulting composite oxide coating included more nickel and less titanium. The prepared NiTi fiber with Ni-rich oxide coating was used to extract typical aromatic compounds coupled with HPLC-UV and exhibited good extraction selectivity for polycyclic aromatic hydrocarbons ( PAHs). The key factors affecting extraction efficiency of PAHs were examined. Under the optimized conditions, the calibration curves were linear in the range from 0. 05 to 500 ng / mL with correlation coefficients ≥0. 999, and the limits of detection were 0. 026-0. 056 ng / mL. Furthermore, the relative standard deviations ( RSDs) for intra-day and inter-day repeatability of the single fiber varied from 4. 8% to 6. 2% and from 5. 4% to 6. 5% for five replicates of PAHs at the spiking level of 50 ng / mL, respectively. The RSDs for the fiber-to-fiber reproducibility of five fibers prepared in different batches ranged from 6. 4% to 8. 4% . This method was suitable for selective enrichment and detection of target PAHs in environmental water samples with relative recoveries of 87. 4% -108. 2% and RSDs <8. 1% . Moreover, this novel NiTi fiber was mechanically strong and chemically stable, and its preparation was precisely controllable.

Objective To explore the effects of water extract from Jiangtang Decoction (WEJTD) on PI3K/Akt signal pathway of skeletal muscle metabolism in KK-Ay diabetic mice.Methods Totally 50 KK-Ay mice were randomly divided into five groups:model group,metformin (positive drug,250 mg/kg) group,WEJTD low,medium,and high dose (2,4,and 8 g/kg) group,with 10 C57BL/6J mice as normal group.The relative drugs were ig administered once a day for 12 weeks,and mice in control group and model group were perfused with distilled water of equal volume.After 12 weeks' oral administration,mice were executed to separate serum,serum insulin level was detected by ELISA kit method;RNA was extracted from muscle tissue by Trizol,and real-time PCR were used to detect the level of PI3K,Akt,GLUT-4,GSK-3β,GS and IRS-1 mRNA.Results WEJTD can down-regulate concentration of insulin in serum and GSK-3β mRNA in skeletal muscle (P < 0.05 and 0.001),and down-regulate mRNA of PI3K,Akt,IRS-1,GLUT-4,and GS in skeletal muscle (P < 0.05,0.01,and 0.001).Conclusion WEJTD decreased glycogen deposition and stimulated glucose transport in skeletal muscle through upregulation of PI3K/Akt signaling pathway.

Objective To study the effect and mechanism of Water extract from Jiangtang Decoction (WEJTD) on diabetes mellitus and diabetic nephropathy (DN) in spontaneous type 2 diabetes mellitus model KK-Ay mice.Methods Totally 50 KK-Ay mice were randomly divided into five groups:model group,metformin (positive drug,250 mg/kg) group,WEJTD low,medium and high dose (2,4,and 8 g/kg) group,with 10 C57BL/6J mice as normal group.The relative drugs were ig administered once a day for 12 weeks,and mice in control group and model group were perfused with distilled water of equal volume.After 12 weeks' oral administration,mice were put into metabolism cages,and the food-intake,water-intake and urine volume were calculated and collected.Blood were collected to detect the concentration of IL-6,ICAM-1 and TNF-α.Then mice were executed,and HE staining and PASM staining were used to check the effect of WEJTD on kidney.Western blotting and qRT-PCR were used to detect the concentration of PI3K,Akt,NF-κB,IL-6,ICAM-1 and TNF-α in kidney.Results WEJTD can alleviate the symptoms of diabetes,such as food ration,polydipsia and polyuria (P < 0.05,0.01,and 0.001);Relief the pathological changes of kidney and significantly decreased glycogen deposition (P < 0.001),down-regulate the increase of IL-6,ICAM-1 and TNF-α in serum and kidney (P < 0.05,0.01 and 0.001),up-regulate the phosphorylation of PI3K and Akt (P < 0.05,0.01,and 0.001),and inhibit the phosphorylation of NF-κB (P < 0.001).Conclusion WEJTD had positive effects on kidney morphology of KK-Ay,and the underlying mechanism might be related to the regulation of PI3K-Akt and NF-κB-mediated inflammation.

RESUMO

A healthy gut consists of commensal flora,epithelial layer and gut-associated lymphoid tissue (GALT). GALT is hyporesponsive to commensal flora and dietary antigens,but can recognize and at-tack pathogens. Accumulating evidence suggests that dendritic cells (DCs) play a crucial role in managing this paradoxical situation and maintaining the complex homeostasis in gut. Influenced by intestinal epithelial cells (IECs) and commensal flora,intestinal DCs possess unique properties that enable them to regulate T-helper 2 (Th2) cells,regulatory T cells (Tregs) and immunoglobulin A (IgA)-producing cells in a steady state. During infection,intestinal DCs are involved in the induction of effector lymphocytes, although they are also responsible for initiating pathogenic responses in inflammatory bowel diseases (IBDs). Therefore, intestinal DCs are associated with not only the maintenance of immune tolerance to commensal flora,but also the induction of protective immune responses against pathogens. This review outlines the roles of commensal flora, epithelial layer, and GALT in mucosal homeostasis and inflammation and summarizes recent progress in DCs-mediated intestinal immune homeostasis.

RESUMO

In the past ten years, the development of electrochemiluminescent ( ECL ) analytical methods based on various types of nanostructures has become a research hotspot. Nanocluster, an intermediate between molecules and conventional nanoparticles, is renowned for its luminescent feature. The first report on ECL for nanoclusters can be traced back to 2009 . Here we summarized the main research progresses since 2011 . Firstly, the preparation of ECL-related nanoclusters was briefly introduced. Then, the mechanisms and applications of ECL by nanoclusters were described. To improve ECL performances, two main strategies, i. e. , nanostructure-based ECL enhancement and biological signal amplification were proposed. Besides, the nanoclusters as the energy transfer receptors in ECL systems were also discussed. In prospect part, the future development of ECL by nanoclusters was considered. We believed that the synthesis of high quality nanoclusters, the reveal of ECL structure-activity relationships, the rationale design and application of near-infrared ECL, and the role of ECL in the interdisciplinary research were the main problems we faced in the future.

Objective To study the effect of urokinase and methylprednisolone combined with psychological intervention on the treatment of the patients with IgA nephropathy. Methods 100 patients with IgA nephropathy treated in our hospital from February 2015 to September 2016 were randomly divided into the control group and the experimental group, with 50 patients in each group. The control group were treated with methylprednisolone, the experimental group were treated with urokinase combined with methylprednisolone and psychological intervention. Pay attention to the psychological state of the, strengthen communication and exchanges, increase confidence and compliance of the patients in the treatment. The therapeutic effects of the experimental group and the control group were compared and analyzed. Results 3 patients in the experimental group were ineffective, and 27 patients were effective. In the control group, 18 patients were ineffective and 24 patients were effective. The effective rate in the experimental group was 94%, which was significantly higher than that in the control group (64%), with statistical difference (P<0.05). After treatment, the level of 24h proteinuria in the experimental group was (0.541±0.322) g / L, significantly lower than that in the control group, with statistical difference (P<0.05). Conclusion The clinical curative effect of urokinase combined with methylprednisolone in the treatment of patients with IgA nephropathy is ideal, which can significantly improve the treatment efficiency, has clinical significance.
